3D細胞活力檢測,需要“極致”手段!

然而傳統的2D細胞活力檢測試劑無法有效的滿足3D細胞活力檢測需求。



今天我們就來看一看,3D細胞活力檢測的“極致”手段。


CellTiter-Glo? 3D 發光法細胞活力檢測試劑盒通過對ATP 的定量來測定3D 培養細胞的細胞活力。ATP 是活細胞新陳代謝的指標。CellTiter-Glo? 3D 細胞活力檢測試劑的特殊配方使其擁有更強勁的裂解能力,專門用于檢測3D 細胞培養產生的微組織(microtissue)的細胞活力,適用于高通量篩選(HTS)。


應用:已成功于多種方式培養的3D 微組織的活力檢測,包括細胞懸滴培養板(hanging-drop plate)、超低吸附細胞培養板、Matrigel 包被的細胞培養板、瓊脂糖包被細胞板、甲基纖維素覆蓋培養液和 Alvetex? 三維細胞培養體系中培養的3D 微組織。

將不同數量的 HCT116 細胞 (RPMI 培養基 +10% FBS) 接種在 InSphero GravityPLUS 96- 孔懸滴培養板, 培養 4 天。 

A. 在微組織培養孔中, 加入與培養基等體積的CellTiter-Glo? 3D 或其他廠家的 A 檢測試劑, 震蕩混勻 5 分鐘后, 室溫孵育 30 分鐘,應用 GloMax? 發光檢測儀讀數。 

B. 2X CellTox 綠色熒光染料 ( 結合于膜完整性受損的細胞的雙鏈 DNA) 提前預混在 CellTiter-Glo? 3D 檢測試劑 ( 左 ) 或 A 檢測試劑 ( 右 ) 中,后共同加入細胞培養孔。 震蕩混勻 5 分鐘后, 室溫孵育 30 分鐘, 應用激光共聚焦顯微鏡拍照。 綠色熒光染色顯示裂解的細胞。 微組織直徑為 300μm。

與不同的3D 細胞培養系統均兼容

HCT116 結腸癌細胞以不同的平臺進行培養:hanging-drop 培養400 個細胞,ULA 和Matrigel? matrix 培養1000 個細胞。生長4 天后,微組織以化合物處理48 小時,然后加入等體積CellTiter-Glo? 3D 試劑,震蕩5 分鐘后,讀取數據。
